Zinc finger proteins, such as ZNF272, interact with nucleic acids and have diverse functions. The zinc finger domain is a conserved amino acid sequence motif containing 2 specifically positioned cysteines and 2 histidines that are involved in coordinating zinc. By screening a human monoblast cell line (U-937) cDNA library with a degenerate oligonucleotide based on a conserved region of Kruppel-related zinc finger proteins that encompasses the H/C link, Abrink et al. (1995) isolated cDNAs encoding 42 distinct Kruppel-related zinc finger proteins, including ZNF272, which they called HZF8. The partial ZNF272 cDNA, which lacks 5-prime coding sequence, encodes 4 functional C-terminal zinc finger domains followed by 1 degenerate finger motif. Northern blot analysis detected a major 7.0-kb ZNF272 transcript in HeLa cells, U-937 cells, and PMA-induced U-937 cells, with higher expression in the monocyte/macrophage lineages.
By large-scale sequence analysis of a fetal brain cDNA library, Dai et al. (2003) cloned ZNF272, which they designated ZNF460. The deduced 562-amino acid protein has a calculated molecular mass of 64 kD. ZNF272 contains a double KRAB domain at the N terminus and 11 C2H2-type zinc finger motifs at the C terminus, suggesting a role in DNA binding. RT-PCR detected ubiquitous but generally weak ZNF272 expression. Highest levels were in pancreas and liver.
Dai et al. (2003) determined that the ZNF272 gene contains 3 exons and spans about 13 kb.
By genomic sequence analysis, Dai et al. (2003) mapped the ZNF272 gene to chromosome 19q13.4.
